How does pKa affect drug distribution?

The pKa of a drug influences lipophilicity, solubility, protein binding and permeability which in turn directly affects pharmacokinetic (PK) characteristics such as absorption, distribution, metabolism and excretion (ADME)1–5.

Why is the pKa important?

Acid dissociation constants, or pKa values, are essential for understanding many fundamental reactions in chemistry. These values reveal the deprotonation state of a molecule in a particular solvent. There is great interest in using theoretical methods to calculate the pKa values for many different types of molecules.

What does the pKa tell you?

The pKa measures how tightly a proton is held by a Bronsted acid. A pKa may be a small, negative number, such as -3 or -5. It may be a larger, positive number, such as 30 or 50. The lower the pKa of a Bronsted acid, the more easily it gives up its proton.

What does high pKa mean?

The pKa scale as an index of proton availability. Low pKa means a proton is not held tightly. pKa can sometimes be so low that it is a negative number! High pKa means a proton is held tightly.

What pKa tells us?

It measures the strength of an acid — how tightly a proton is held by a Bronsted acid. The lower the value of pKa, the stronger the acid and the greater its ability to donate its protons.

What determines pKa?

The strength of the acid (pKa) depends on the stability of the base. When the proton leaves the acid, it leaves behind its electrons. Those are super negative, and there is a big negative charge on the conjugate base. You know that if there is a concentrated negative charge, the base is not very stable.

Does a high pKa mean more acidic?

In addition, the smaller the pKa value, the stronger the acid. For example, the pKa value of lactic acid is about 3.8, so that means lactic acid is a stronger acid than acetic acid.

What happens at pKa?

When the pH equals the pKa, the acid molecules are 50% protonated and 50% deprotonated. At a pH below the pKa, more acid is protonated.
