What does BRAF kinase do?

BRAF protein is a serine-threonine kinase that is encoded on chromosome 7q34. It is an important signal transduction molecule that mediates signals from RAS to MEK, ultimately promoting cell proliferation and mobility.

What is a BRAF protein?

BRAF is a gene found on chromosome seven that encodes a protein also called BRAF. This protein plays a role in cell growth by sending signals inside the cell promoting, among other functions, cell division.

Does BRAF phosphorylate MEK?

In such a model, MEK1 acts as a scaffolding protein to stabilize the active conformation of BRAF and aligns the activation loop for auto-phosphorylation. Subsequent to auto-phosphorylation, fully activated BRAF phosphorylates MEK1 to relay downstream signal.

What is BRAF testing?

A BRAF genetic test looks for a change, known as a mutation, in a gene called BRAF. Genes are the basic units of heredity passed down from your mother and father. The BRAF gene makes a protein that helps control cell growth. It’s known as an oncogene. An oncogene works like a gas pedal on a car.

What does BRAF positive mean?

“BRAF positive” means that your tumor has a mutation in the BRAF gene. This gene controls a protein that stimulates cell growth. When there’s a mutation, it causes the continuous production of this protein, which can lead to unchecked cell growth or cancer.

What does a positive BRAF test mean?

A BRAF mutation is a change in a BRAF gene. That change in the gene can lead to an alteration in a protein that regulates cell growth that could allow the melanoma to grow more aggressively. Approximately half of melanomas carry this mutation and are referred to as mutated, or BRAF positive.

How does V600E activate BRAF?

Activating mutation in BRAFV600E activates the MEK/ERK pathway in the absence of extracellular signaling. Activating mutations in RAS can stimulate MEK/ERK signaling through CRAF in the absence of extracellular signaling. BRAF and RAS mutations occur in a mutually exclusive fashion.

Is BRAF positive good?

Getting a positive test for the BRAF V600 tumor mutation is certainly good news for a newly diagnosed melanoma patient. This is because some targeted drugs work very well against such tumors, which account for about 50% of melanomas.

What is v600e BRAF mutation?

myoo-TAY-shun) A specific mutation (change) in the BRAF gene, which makes a protein that is involved in sending signals in cells and in cell growth. This BRAF gene mutation may be found in some types of cancer, including melanoma and colorectal cancer. It may increase the growth and spread of cancer cells.

BRAF testing is done to look for genetic changes in tumors (genomic alterations) that are present in some cancers, including metastatic melanoma, lung cancer, colon cancer, and others. If positive, the presence of a BRAF mutation may help guide treatment (such as medications that target BRAF mutations), estimate prognosis, and more.

What is a body biopsy BRAF test and how is it done?

BRAF testing is most often done on a sample of tissue taken during a biopsy or removal of a tumor. This can pose challenges, however, as biopsy procedures to obtain tissue are invasive, and even when done, there are times when there is not enough tissue present to do the testing.

What are the risks of BRAF testing?

A with any laboratory test, BRAF testing is subject to lab error. The primary risk of BRAF testing is the procedure used to obtain the sample. This can vary depending on the type of cancer, the site of the cancer, the general health of a person, and more.

What should I do if my BRAF test is negative?

If BRAF testing is negative on tissue testing (depending on the type of your cancer), a liquid biopsy may be considered (or vice versa). Likewise, if BRAF testing is negative on a rapid test, comprehensive gene profiling may be considered. Tumor Progression and/or Spread
